Optimization of storage conditions for Puumala virus based vaccine

چکیده
We report the effects of storage conditions and human serum albumin addition to vaccine composition on immunogenic properties an experimental against hemorrhagic fever with renal syndrome. To obtain vaccine, Puumala virus, propagated in Vero cells culture, was concentrated, inactivated beta-propiolactone, purified by chromatography CaptoCore 700 multimodal sorbent (GE Healthcare). The target component 20.2 106 viral RNA copies/ml. humoral immune response determined measuring neutralizing antibodies blood immunized Syrian hamsters (Mesocricetus auratus). It revealed that virus beta-propiolactone induces a pronounced response, which indicates preservation relevant epitopes. Аccording our study, is more stable during vs. intact virus. confirmed full inactivation occurred after 24 hours at 37C. A significant decrease immunogenicity thermal noted, confirms need for careful selection thermolabile Hantaviruses. direct correlation observed between titer copy number/ml. Adding stabilized infectivity storage: remained same level 3 months 62C, whereas without albumin, decreased undetectable level. 0.1% its long-term storage. shown preparations (0.11%) show higher dose-dependent stability upon repeated freezing assessed detecting their immunogenicity. can be assumed provides better genetic material storage, as well protein epitopes responsible protective form induction, undergo less conformational changes presence albumin. These data support formulation.

Optimization of conditions for gene delivery system based on PEI
Objective(s): PEI based nanoparticle (NP) due to dual capabilities of proton sponge and DNA binding is known as powerful tool for nucleic acid delivery to cells. However, serious cytotoxicity and complicated conditions, which govern NPs properties and its interactions with cells practically, hindered achievement to high transfection efficiency. Here, we have tried to optimize the properties of ...
متن کاملEnvironmental conditions and Puumala virus transmission in Belgium
BACKGROUND Non-vector-borne zoonoses such as Puumala hantavirus (PUUV) can be transmitted directly, by physical contact between infected and susceptible hosts, or indirectly, with the environment as an intermediate. The objective of this study is to better understand the causal link between environmental features and PUUV prevalence in bank vole population in Belgium, and hence with transmissio...
متن کاملConstruction and nonclinical testing of a Puumala virus synthetic M gene-based DNA vaccine.
Puumala virus (PUUV) is a causative agent of hemorrhagic fever with renal syndrome (HFRS). Although PUUV-associated HFRS does not result in high case-fatality rates, the social and economic impact is considerable. There is no licensed vaccine or specific therapeutic to prevent or treat HFRS. Here we report the synthesis of a codon-optimized, full-length M segment open reading frame and its clon...
